This study focuses on the stimulation and protection of entrepreneurship in the construction of "three regions and one district" in Anhui Province. Through in-depth analysis of the policy environment, regional economic development, social and cultural environment factors on the influence of entrepreneurship, and discusses the key path to stimulate the entrepreneurship and protect the formation mechanism, combined with the practice of Hefei high-tech industrial development zone, this paper reveals the entrepreneurship in promoting the core role of regional economic development. The study found that the policy guidance, the demonstration of excellent entrepreneurs, a cultural atmosphere of tolerance for failure and a diversified support system are the key factors to stimulate and protect entrepreneurship. Based on these findings, this paper presents targeted policy recommendations and prospects future research directions. By stimulating and protecting the entrepreneurial spirit, it can further promote the vigorous development of the "three regions and one district" construction in Anhui Province, and also provide useful reference for other regions.
